Clinical relevance

Benzodiazepines have a broad range of applications and are the main representatives of sedatives and sleeping pills. Some are also used as antiepileptic drugs, muscle relaxants, and as analgesics in administering general anaesthesia. In the case of alcohol withdrawal treatments they facilitate weaning, although the danger of an alternative addiction exists. The various benzodiazepines are metabolised at different rates, and in some cases it is not until the metabolites form (e.g. desalkylflurazepam) become therapeutically effective.

Regular determination of the plasma levels minimise the risk of side effects resulting from overdose. Furthermore, patient compliance can be confirmed to help ensure optimal therapy outcomes.

This Chromsystems kit allows for the easy and reliable chromatographic determination of a total of 15 benzodiazepines and metabolites. Sample preparation is based on an efficient and selective solid phase extraction, which can also be automated with the Gilson® ASPEC™. Chromatographic measurement is performed on an isocratic HPLC system with UV detection.

Pre-analytic Treatment

Patient specimens are stable up to 1 week stored at +2 to +8 °C. For longer storage periods deep-freeze the patient specimens below -18 °C.

Sample Preparation


  • Condition the Sample Clean Up Columns with 1 ml Equilibration Buffer 1 and 2 each.
  • Apply 1 ml plasma/serum and 100 µl Internal Standard, mix several times.
  • Draw through completely by centrifugation (1 min at 340 x g) or suction, discard effluent.


  • Apply 2 x 1 ml Wash Buffer to the Sample Clean Up Column and draw through by centrifugation (1 min at 340 x g) or suction, discard effluents. Then centrifuge 2 min at 1090 x g to dryness or use suction.


  • Apply 400 µl Elution Buffer to the Sample Clean Up Column, draw through completely by centrifugation or suction, then dilute with 300 µl distilled water.
  • Inject 50 µl of the diluted eluate into the HPLC system.

Sample Stability

At +2 to +8 °C up to 1 week. Freeze for longer storage.
